Key steps of the biosynthesis of the polyketide antibiotic mupirocin have been demonstrated using assays that mimic the biosynthesis in-vitro. Fatty acid construction in mupirocin was shown to proceed using two ketosynthase domains (MmpF and MmpB_KS) and a series of other tailoring domains. The acyl carrier protein has been shown to play a vital role in shuttling the correct intermediates to the active site of each protein.
